AmilCP screening part

This part has a strong constitutive promoter (J23100) the standard RBS (B0034) and AmilCP (K592009). When assembled into a plasmid it gives a strong blue color.

The reason we created this part is the default backbones all have essentially the same construct but using RFP instead. Using an insert with a different color it made it much easier to debug problems with digests, ligations and transformations. We also used it to try to find a problem with some strange background issues we had.

This part is also very useful for protein purification experiments. In a standard 0.5% agarose gel it has an advantage of running up on the gel while RFP runs down on a gel. AmilCP absorbs at 588 nm while RFP excites at t 584 nm and emits at t 607 nm. However RFP has a secondary absorption peak at 502 nm and this is far enough from the 588 nm peak for AmilCP that it works extremely well. This allows AmilCP and RFP to be mixed and separated with an easy verification to figure out how effective the separation was.
