We first construct tandem double promoter to prove our ideas appliable. We use consecutive promoters J23102, J23106 and J23116 to produce seven different combinations of double tandem promoters(figure1) .The double tandem promoters provide several different dcas9-guide RNA target sites,by which we could achieve multiple-stage modulation of the target genes. We use single promoter J23106 as a quantified control to check the strength of the tandem double promoter. The strength of the double promoter BBa._K1081006 is .
