The bacterial cultures were grown for 12 hours in LB Media containing appropriate antibiotics and centrifuged at 13000 RPM for 1 minute. The cell pellets for all three cultures were analyzed using UV filter for GFP expression.

Flow cytometry was carried out on bacterial cultures expressing inverted GFP and bacterial cultures expressing GFP (BBa_I20260). A shift in the fluorescent population was seen for both cultures when compared to a bacterial culture expressing no fluorescent protein. This shift in fluorescent populations for both GFP and the modified GFP (GFP with an ORF in a reversed orientation compared to convention) were similar, which confirms similar expression from both GFPs.
