The hygromycin phosphotransferase gene (hph), which also confers resistance to hygromycin, sits downstream of the T7 promoter; this construct can be used as a selectable marker in co-transformation with T7 polymerase in fungi.
Usage and Biology
- The part was constructed from BBa_I712074 and BBa_K1021002.
- Cornell iGEM characterized this part in E. coli BL21-A1 using a zone of inhibition test. The data is displayed below; at concentrations of 200 and 2000 μg/mL, the wild-type strain was significantly more inhibited than the allegedly hygromycin-resistant strain (one-tailed student's t-test, α=0.005).
