EN-Ptet-rbs-X-[GFP]-SNP "ORF Expresser"

Plasmid for assembly of open reading frame basic parts. A Tet promoter (r0040) and strong ribosome binding site (b0034) lie between the EcoRI and XbaI sites. GFP (just the ORF, E0040) lies between XbaI and SpeI. The part can be used two ways:

1. Insertion of a standard ORF part into the XbaI/PstI sites results in a Ptet/RBS driven protein generator while retaining the ability to do biobrick assembly.
2. Insertion of a promoter/RBS cassette (cutting the part with EcoRI/SpeI, inserting into EcoRI/XbaI of J61003) results in a standard pSB1A2 plasmid containing an your promoter/rbs.E0040 composite.

Part J23018 is J61003 with RFP instead of GFP. Part J23019 does the same thing with RFP in a pSB3C6 plasmid backbone. See similar part J61002 for a method of construction for promoter parts, or r0040.part composite parts in pSB1A2.
