Design Notes
We considered two designs for exponential phase regulation of the wintergreen odor enzyme generator: an exponential phase promoter or a stationary phase promoter in combination with a transcriptional inverter device. Both designs require a functional, growth phase dependent promoter. The unique challenge in using an exponential phase promoter would be coordinating the timing of the exponential phase regulation of wintergreen odor and the stationary phase regulation of banana odor. The unique challenge in using a stationary phase promoter in combination with an inverter is that functional composition of transcriptional devices often leads to unpredictable device operation. Thus, the transcriptional control device could fail to produce a signal during exponential growth or could fail to turn off during stationary phase. Our reuse of an "off-the-shelf" transcriptional inverter from the Registry saved us considerable effort in constructing an exponential phase control device.
