BBa_J434007 was assembled into a genetic construct using Golden Gate Assembly, allowing for the integration of the terminator in a precise location within the construct, downstream of the eYFP reporter gene. The fluorescence data collected during the experiment, using a plate reader, shows the efficiency of BBa_J434007 in terminating transcription under ambient conditions (28°C). According to the terminator activity graph, BBa_J434007 demonstrated a steady increase in activity during the first half of the time course. Following this peak, the activity began to decline gradually over the remaining time, though it remained relatively high compared to earlier time points, suggesting it may perform well under ambient temperature conditions but may require further analysis to understand its behavior over extended periods.
