BBa_B0034_m1 was assembled into a construct using Golden Gate Assembly, with eYFP as a reporter to measure RBS activity via fluorescence under two temperature conditions. BBa_B0034_m1 demonstrated strong and consistent activity at both 28°C and 37°C, as shown by the fluorescence data. At 28°C, the RBS maintained stable high performance throughout the time course, while at 37°C, it peaked early and remained fairly constant over time. These results indicate that BBa_B0034_m1 is a highly efficient RBS suitable for use under both ambient and heat stress conditions.
