Design Notes

Polylinker had to be set up in a manner to facilitate compatible adjacent double digests. Also, polylinker sites had to be completely unique to vector sequence.

Plasmid comes from pBR322, the inserted polylinkers was a custom construct of two annealed oligonucleotides from invitrogen.
