Sample 1 is the positive control induced with light; sample 3 is the positive control with no light; sample 5 is experimental 1 with no light; and sample 7 is experimental with no light; and sample 9 is simply water. The positive control was exposing the cells to light; the negative control was keeping the cells in the dark; and the experimental was the insertion of the promoter into the cells. No cells were present in the samples where the promoter was inserted and so sample 9 with water is used the represent the lack of cells. Because both the positive control induced with light and the positive control with no light exhibited fluorescence, the promoter does not function as a light-inducible one.
