cis4-repressed, tet-regulated Q
This part has the Q construct from I759003 with the cis repressor cis4 cloned in to replace the spacer. The weak constitutive promoter J23116 will produce a steady stream of tetR, which (in the absence of aTc) repress transcription of the pTet-cis4-Q transcript. The riboregulator design further represses levels of Q protein in the cell. The transcript will form a cis-hairpin loop and prevent translation in the absence of a riboregulator trans sequence, which binds to and opens up the cis-hairpin, allowing the ribosome to access the RBS site.
