Grew E.coli colonies in LB media overnight. Positive control contains working pScarlet promoter. Negative control contains LB media with ampicillin. Experimental 1-3 were extracted from colonies transformed with newly designed pScarlet promoter. We measured fluorescence and absorbance of each sample. Bars display ratio of fluorescence to absorbance, adjusted so that LB media had a baseline of zero (e.g. fluorescence= sample-LB media). Negative and experimental 3 showed negative values, which we converted to zero.
