Submission Format

As in previous years, parts must be BioBrick compatible and in pSB1C3.


Additionally, all part submissions must be:

  • miniprepped plasmid DNA (over 250ng) dried down. 10ul of 25ng/ul for a total of 250ng works well.
  • in 96 well plate format (label your plate with your team name and submission number)
  • plate sealed with adhesive foil
    • the adhesive foil should make full contact with the wells of the plate (see picture)
  • sealed plate covered with a plastic lid
  • shipped with a tracking number
  • shipped and labelled as DNA, not hidden or disguised
    • DNA (non-hazardous, non-regulated, non-infectious, for research purposes only)

Submission Kit Contents

  • (3) 96-well skirted PCR plates
  • (3) universal plate lids
  • (5) adhesive foils
  • bubble wrap
  • a box with the iGEM HQ address, for shipping your submission




Sample Layout

Make sure to add your samples in the correct layout:

  • Your first sample (s1) should be dispensed into well 1A of your submission plate.
  • Your second sample (s2) should be dispensed into well 1B of the same submission plate.

Use one plate per a submission order. Example: If you are submitting 104 parts, create one submission order for 96 parts and another submission order for 8 parts. Label the plates with the correct submission order.



Submission DNA Preparation

  • 250ng minimum of purified plasmid DNA (your BioBrick compatible part in pSB1C3)
    • 10ul of a 25ng/ul miniprep will work well
  • DNA must be dried down before shipping

We recommend drying down the plate in your lab's laminar flow hood (you can also leave your plate on the lab bench to air-dry, covered with the plastic lid).

  1. Sterilize the hood surface with 70% Ethanol
  2. Cover the 96 well plate with the provided plate lid (not with the adhesive foil)
  3. Place the plate in the hood.
  4. With sufficient airflow, drying down 10ul of 25ng/ul (250ng total) of purified plasmid DNA in the plate should take 4-6 hours (or overnight).
  5. Once all wells have dried, cover the plate wells with the provided adhesive foil.
    1. The adhesive foil should make full contact with the wells of the plate (see picture). When properly sealed you should be able to see the impression of every well.
  6. Cover the sealed plate with a plastic lid
  7. Label your plate




Plate Labelling

Make sure to label each plate with your group (team/lab) name and the appropriate submission number. Use a lab marker and write your team name and submission number on the side of the 96w plate.

Use one plate per a submission order. Example: If you are submitting 104 parts, create one submission order for 96 parts and another submission order for 8 parts. Label the plates with the correct submission order.

Note: When filling shipment info please make sure to declare and label the package as "DNA (non-hazardous, non-regulated, non-infectious, for research purposes only)."


Packaging your submission for shipping

Place your submission in an appropriately sized shipping container, such as a cardboard box, along with the printout of the submission form. If you use cushioning material inside the container, limit cushioning material to foam peanuts, bubble wrap, packaging paper, or similar.

Note: Part submissions packaged in materials other than the above will be rejected and these part submissions will not be taken into consideration for medals or awards. It is important that you not hide your submissions in an effort to conceal them from customs, because they will be seized.
