pucpro+GFP

puc promotor region of R. sphaeroides (normally promotes production of LHII antennae (pucBA, as well as C,D,E)), and green flourescent protein


Usage and Biology

The puc promoter is the primary promoter for the puc operon. The puc promoter is ultimately controlled by oxygen tension, and is a weak promoter at high oxygen tension (200 uM dissolved O2) and a very strong promoter under low oxygen tension (10 uM or less dissolved O2). At intermediate oxygen tension (100 uM dissolved 02) expression from the puc promoter is dependent on light intensity such that expression is downregulated under higher light intensities. This strict mechanism of control is thought to reflect the potential for photodamage as purple bacteria do not have well-evolved systems for dealing with reactive oxygen species.
