Usage and Biology

Photorhabdus virulence cassette (PVC) is one group of extracellular Contractile injection systems (CISs) that are cell-puncturing nanodevices for targeted delivery of proteins .[1]Structurally similar to other CISs, PVCs are also macromolecular complexes containing a rigid tube structure housed in a contractile sheath, which is anchored to a baseplate and sharpened by a spike protein.[2]

The PVC tube linked with payloads consists of three structurally similar tube proteins, such that a complete syringe in a typical length contains 24 stacked hexameric rings of tube proteins, including 2 layers of tube initiators and 22 layers of main tube proteins. Among all related genes,pvc1 produces pvc1 hexameric rings which are assembled to form main tube proteins through opposite surface charges and electrostatic attraction.As a whole,the inner surface of Pvc1 tube is remarkably negatively charged for loading its payloads.
Furthermore,pvc1 interacts with the part of sheath comprises alternating Pvc2 and Pvc3 subunits ,which appears to be dictated by electrostatic interactions as well.This aims to prepare for the contraction in the delivery process.
