The lac operator used in pET28a is a specific DNA sequence derived from the lac operon of Escherichia coli. It serves as a regulatory element in the plasmid's gene expression system. The lac operator is the binding site for the Lac repressor protein (LacI). When LacI binds to the operator, it blocks the transcription of genes downstream of the operator, including the T7 promoter that drives the expression of the target gene in pET28a.
