Design Notes
It is necessary to design DNA sequences with appropriate length to produce mRNA binding switches according to the types of different proteins. Taking human insulin proteinogen as an example, we hope to use this switch of mRNA interaction to ensure the normal growth of the strain without expressing the downstream killing system when the expression level of insulin mRNA is normal, and to start the expression of the downstream killing system and kill the deceiver when the mutation at the key site of proteinogen gene leads to the abnormal expression level of its mRNA. At the same time, we hope to explore the development possibility of mRNA interaction switch, and explore the optimal length ratio and optimal conditions of two mRNA interactions.
