Introduction
The HygR cassette is composed by the promoter ADH1, the resistance gene hygR and the transcription terminator of VPS13. The resistance gene is based on the part (BBa_K2386004), but it was codon optimized for expression into S. cerevisiae.
This gene codes for a hygromycin phosphotransferase, which inactivates the hygromycin by adding a phosphate on it. Therefore, this cassette allows for the selection of yeast transformants upon addition of hygromycin into the media of culture.
Part characterization
The part BBa_K3930011 was used to select the genomic integration of the part (BBa_K3930000). Transformants were plated onto YPD + 200 μg.ml-1 hygromycin Petri dish. The Toulouse INSA UPS team managed to select transformants with this selection marker (for more details, check the BBa_K3930000 part page).
