T7-GamS-HisTag

This is a composition of the coding sequence for the protein GamS(BBa_K2203000) with a T7 promoter and terminator as well as a 6X His-tag for easy transcription and purification of the GamS protein. GamS is a protein that protects linear DNA templates from degradation in a cell-free environment. It is used to enhance cell-free protein synthesis by inhibiting exonuclease activity. For a detailed description and experimental demonstration of the effect of GamS on cell-free protein synthesis, please refer to the page of GamS (BBa_K2203000). This composite part includes a T7 promoter and terminator for enhanced transcription of GamS-6XHis during growth of bacteria transformed with the plasmid containing this part. GamS can then be specifically purified or detected in the cell lysate by making use of the 6XHis tag.
