In the "pBad" construct, when the pBad promoter (green bent arrow) is inverted, the restriction fragment changes from 200 bp to 300 bp. In the "Tet" construct, when the RBS-Tet coding region is inverted, the restriction fragment changes from 200 bp to 1000 bp.
