Three cloned GBS were ligated to pSB1C3 plasmid backbone. One control pSB1C3 plasmid was taken with a RFP coding insert. Four shake flask cultures were set up and the mini-prep DNA samples were run on chloroquine gels to compare the level and quality of supercoiling in the plasmid
