The Chiba iGEM team tested this part in several conditions.
- Without LuxI (or synthetic OHHL), RFP levels increase.
Experiment : checking BBa_I731012
- Picked and cultured the following stab and glycerol stocks in 2mL of LB:
- LB-Amp, BBa_I731012, (stab, received from iGEM)
- LB-Amp, BBa_K084007(plac+rbs+LasI(no LVA)), (JW1908)
- LB-Amp, BBa_K084012(plac+rbs+LuxI(no LVA)), (JW1908)
- Pre-Cultured at 37°C for 12h.
- Added 6.25% each of the pre-cultures to new LB medium.
- LB-Amp, BBa_T9002, BBa_K084012(plac+rbs+LuxI(no LVA)), BBa_K084007(plac+rbs+LasI(no LVA))
- Cultured at 37°C for 12h
- Mixed the sender cells BBa_K08400 and 7BBa_K084012 both with BBa_I1731012 at a 1:1 ratio.
- BBa_I1731012 alone.
- BBa_K084007:BBa_BBa_I1731012=1:1
- BBa_K084012:BBa_I1731012=1:1
- Culture 12h and observe results(follow picture).
- Funakoshi, UVGL-58 : long wave(365 nm)
