Promoter plac

plac is an operon sensitive to LacI protein and CAP factor. We used the plac operon in a controlled trial.


Usage and Biology

The plac is derived from the E. coli lactose operon and consists of a repressor protein gene (LacI), a promoter gene (P), a operon gene (O) and a gene structure encoding three enzymes related to lactose utilization. Its positive regulation activates the promoter through CAP factors and cAMP to induce transcription. Negative regulation is through the production of LacZ blocking protein by the regulated gene. This repressor protein binds to the manipulated gene to block transcription. Lactose and some analogues, such as IPTG, can form a complex with the repressor protein to unblock it and induce transcription to occur. In our experiments, we used the plac promoter as a control and fused it to LuxCDABC (BBa) for observing the change in abundance of the strain.
