UCSF-UCB iGEM 2014

We used pTEF1 in pSV606 (S. cerevisiae) yeast strains to create a constitutive circuit for our project, and it performed consistently as a constitutive promoter. We characterized pTEF1 alongside some of its mutants (m3, m6, m7, and m10), as seen in the following graph. If one would like to optimize expressiveness, pTEFm7 was found to have a higher level of constitutiveness than pTEF1, but pTEF1 is still a reliable promoter.
