Registry groups must follow iGEM's submission prep guidelines.
Registry's Submission System
The Registry's submission system gives feedback to users and helps the Registry process new submissions. You'll see when your physical shipment has arrived at the Registry, and how your samples are being evaluated on an individual basis. If there is a mistake with the form, or an issue with a sample, you won't have to redo your submission form.
You've added your part to the Registry, but before you send a DNA sample to the Registry, make sure that your part follows the Registry's submission requirements and instructions.

pSB1C3
- Parts must be in pSB1C3, the Registry's standard shipping backbone
- Your part will be flanked by the BioBrick prefix and suffix on the backbone
- The Registry can test and maintain all parts in the same way
- pSB1C3 is a high-copy plasmid, which improves miniprep yields for parts
- Users can easily move/assemble the part into another plasmid backbone
- You can use the pSB1C3 linearized plasmid backbone (in kit or by request) or the pSB1C3 samples in the distribution kit.
BioBrick compatible
- Parts must be BioBrick (RFC 10) compatible
- Your part's sequence must not contain the restriction sites of the BioBrick prefix and suffix: EcoRI, XbaI, SpeI, PstI.
- The Registry can test and maintain all parts in the same way
- Your part can be assembled to others with 3A Assembly.
- Users can easily move/assemble the part into another plasmid backbone
- Check if your part is BioBrick compatible by looking at the part's sequence and feature box.
Documented
- Parts shipped to the Registry must be documented on the Registry.
- You must add your part to the Registry; it will then receive a part name (BBa_...)
- Your part's sequence must be entered into the Registry
- Quality over quantity: sending a few well-documented parts is always better than sending several that are not

Safety
- iGEM teams must meet the safety requirements
- The Registry accepts parts from Risk Group 1 organisms.
- The Registry may accept parts from Risk Group 2 organisms that have received authorization from the Safety committee
- The Registry will not accept any parts from Risk Group 3 and 4 organisms
- The Registry will not accept any parts from organisms or viruses on The Australia Group's List of Biological Agents for Export Control.
- Now that your part meets the Registry's submission requirements, you're ready to send a DNA sample:
- you'll need to fill out an online submission form
- prepare the physical shipment (aliquot DNA into tubes or wells)
- ship the DNA to iGEM HQ (include a tracking number)
Note to iGEM Teams: Your BioBrick Part DNA is due at iGEM HQ by the deadline on the Calendar of Events. Make sure to ship it before the deadline, allowing for transit time.

- BioBrick a part: add the BioBirck prefix and suffix to your part through PCR
- Linearized Plasmid Backbones: transfer your part into the pSB1C3 linearized plasmid backbone
- Use site mutagenesis to remove illegal restriction sites
