Luminesensor repressible recA408 promoter

RecA promoter in 408 form.
RecA promoter belongs to the SOS regulon family. In E.coli it promotes the transcription of RecA protein, which forms filament when bound to ssDNA and trigger the self-cleavage of LexA protein. LexA protein can also bind to RecA promoter, inhibiting the transcription of the genes downstream, too.
However, with mutations in specific sites which LexA protein will bind to, RecA408 promoter will not be bound by LexA protein, but is compatible to LexA408 protein with corresponding mutations in DNA binding site. So a bio-orthogonal promoter is formed because the influence of endogenous LexA protein is excluded.
For example, the optimized Luminesensor with a LexA408 domain (BBa_K819005) constructed by [http://2012.igem.org/Team:Peking Peking 2012 iGEM team] can specifically bind to recA408 promoter and therefore inhibit the gene expression (e.g. BBa_K819007, a measurement device containing RecA408 promoter constructed by Peking 2012 iGEM team)
