Plux (rep) R0061middle
R0061 is the promoter repressed by LuxR in the presence of AHL. We designed this promoter by changing one base of the existing BioBrick (BBa_R0061).
In order to characterize this promoter, we constructed K395105 combining K395008 and K121013, which is a promoter-less gfp reporter (rbs-gfp-ter-ter) on pSB6A1. In order to measure activity of R0061, we introduced K395105 and S03119 (pTetR-rbs-luxR-ter) on pSB3K3. The left figure shows that this promoter is repressed by 3OC6HSL and the strength of this promoter is weaker than R0061.(→[http://2010.igem.org/Team:Tokyo_Tech/Project/Artificial_Cooperation_System/lux_act_rep#R1, characterization of R0061 & K395008 (promoter repressed by LuxR/3OC6HSL) more information])
