The Zif268 zinc finger sequence was ligated into an existing BioBrick vector containing the Elowitz RBS site (Part:BBa_B0034). The vioB gene was obtained by PCR from an existing BioBrick of violacein operon (Part:BBa_K274002) and ligated as a back insert into the RBS_zinc-finger construct.

The vioB sequence was obtained by PCR from an existing part, which was designed by the Cambridge 2009 iGEM team (Part:BBa_K274002). The Zif268 zinc finger is a synthetic gene, which our team designed and ordered from GeneArt.
