This part is composited from the Blues zinc finger gene(Part:BBa_K323136) and the ''vio''A gene, which codes for the first enzyme in the violacein biosynthetic pathway. The resulting proteins are connected with a glycine serine linker. Therefore, the product of the DNA sequence is a chimeric VioA enzyme, which binds to a specific DNA sequence.

Design Notes

The Blues zinc finger sequence was ligated into an existing BioBrick vector containing the Elowitz RBS site (Part:BBa_B0034). The vioA gene was obtained by PCR from an existing BioBrick of violacein operon (Part BBa_K274002) and ligated as a back insert into the RBS_zinc-finger construct.

The vioA sequence was obtained by PCR from an existing part, which was designed by the Cambridge 2009 iGEM team (Part:BBa_K274002). The Blues zinc finger is a synthetic gene, which our team designed and ordored from GeneArt.
