Design Notes
We designed the signal peptide by taking the verbatim signal peptide from the beta-lactamase gene of ATCC 12228, a gene that we know is targeted for secretion by the cell through the Sec pathway. We searched for the sequence in the signal peptide database http://proline.bic.nus.edu.sg/spdb/. We then had the sequence synthesized by GeneArt(TM) and made into a biobrick standard part.
