Design Notes
We mainly consider the role of nadK in changing intracellular NAD+ and NADP+ levels. Given the NADPH requirement of the engineered bacteria, we decided to introduce NADK encoded by nadK. NADK can phosphorylate NAD and produce NADP(H). NADK is the only known mechanism by which NADP(H) is produced de novo.
