In composite parts assembled according to assembly standard 37 KSI-DP basic part is added to other parts by cutting front inserts with EcoRI and SpeI and front vectors with EcoRI and XbaI, thus forming scar Xba/Spe NgoMIV (BBa_K245080) beween KSI-DP and another part. This is necessary since BB-NIC-III-HisN vector containing KSI-DP (BBa_K245113) does not contain NgoMIV and BspEI restriction sites in its multiple cloning site (which were usually used to assemble composite parts). There is additional NgoMIV restriction site in the scar since BB-NIC-III-HisN vectors containing other parts (iGEM 2009, team Slovenia) have NgoMIV and BspEI restriction sites in their multiple cloning site (XbaI-NgoMIV-part-BspEI-SpeI).

This is necessary since BB-NIC-III-HisN vector containing KSI-DP (BBa_K245113) does not contain NgoMIV and BspEI restriction sites in its multiple cloning site (which were usually used to assemble composite parts). There is additional NgoMIV restriction site in the scar since BB-NIC-III-HisN vectors containing other parts (iGEM 2009, team Slovenia) have NgoMIV and BspEI restriction sites in their multiple cloning site (XbaI-NgoMIV-part-BspEI-SpeI).
