3' 20 nt overlapping region for pUC19 EcoRI site
We incorporated overlapping regions into each insert because we're using the HiFi assembly method for assembling all the insert parts in our project. The pUC19 vector was digested and linearized using EcoRI and PstI restriction enzymes.
We also designed a 20 nt overlapping region at the 5' end for the PstI site on the pUC19 vector (BBa_K5152010).
We verified that these regions allow for successful and effective assembly.
