The gene acaP comes from Rhodopseudomonas palustris CGA009, and it can encode a kind of α-carbonic anhydrase that is able to speed up the reversible conversion between CO2 and bicarbonate. In our design, engineered Pseudomonas aeruginosa PAO1 converse CO2 into bicarbonate, in order to deliver the carbon source in the biofilm more easily. Rhodopseudomonas palustris CGA009 take in the bicarbonate and transform it as CO2 for CBB cycle. The efficiency of the conversion in Rhodopseudomonas palustris WT is limited. Therefore, we introduce acaP into Rhodopseudomonas palustris CGA009 to realize overexpressing of the carbonic anhydrase, which enable the engineered Rhodopseudomonas palustris to converse bicarbonate into CO2 in a higher rate, accelerating the process of carbon fixation in Rhodopseudomonas palustris CGA009.
