araBAD-weak-B0034-ccdB

Biology

pBAD/araC promoter consists of two structures, namely araC and araBAD promoter. We found that BBa_K206001 and araBAD promoter part overlapped, so we tried to combine BBa_K206001 with the unoverlapped part to build an araBAD-weak promoter.

We constructed composite part:BBa_K4907141.

Characterization

Colony PCR

In the construction of this circuit, colony PCR and gene sequencing were used to verify the correctness of the transformants. At a position around 916bp, a target band of approximately 1000bp was observed (Fig. 2).
