Design Notes

"1. AraI is selected while AraO2 is not to explore the oppotunity to design a NOT gate. The arabinose binding condtion of AraC will bind to AraI, which enables us to directly try to use it as a repressor instead of using the original DNA loop regulation. 2. Double AraI are utilised to explore the oppotunity of increase the repression effect. 3.T-rich spacers results in nucleosome-free regions to reduce the potential impacts from neighbouring DNA.
