Biology and Usage
The helper plasmid is part of a system to create ssDNA using bacteria. This can be used for DNA origami.
When using the helper-plasmid approach, the pScaf containing the DNA-origami sequence (BBa_K4665170) and the helper plasmid are both introduced into the bacteria by simultaneous transformation. The helper plasmid contains the origin of replication to be able to produce the ssDNA sequence needed for DNA origami.
The helper plasmid was constructed by Praetorius et al.,(2017) by cloning the coding parts of helper phage M13KO7 into a pSC101 plasmid backbone carrying a kanamycin resistance gene.
