We used pUAST vector system to construct pUAST-MTF-1 plasmid. pUAST vector contains UAS and Hsp70. We inserted MTF-1 gene in the pUAST vector downstream of UAS and Hsp70 by enzyme cutting and linking, and constructed pUAST-MTF-1 plasmid which was then transfected into Drosophila S2 cells. We also inserted GAL4 gene into the cells. GAL4 binds to UAS and can activate the expression of MTF-1 gene downstream of UAS-Hsp70.
