Design Notes

MazF can be characterized in a cell density-dependent manner in E. coli. When the expression of both MazF and MazE genes is turned off (because both are constrained by the same promoter), MazE degrades faster than MazF. If there is nothing to inhibit MazF, then the toxins produced will eventually kill the cells.


Source

The tetracycline repressor(tetR) is derived from transposon Tn10 (+LVA). The lacI repressor and T7 terminator are derived from E. coli (+LVA). The MazF and MazE are derived from Bacillus subtilis.
