This part was used by Edinburgh-UHAS_Ghana team of 2022 to test cell-free transcription protocols. The construct contains an F30 flanked Squash aptamer (BBa_K4390096) under a strong T7 RNA promoter (BBa_I712074). When used in a cell-free transcription system, the construct is trancscribed by a T7 RNA polymerase constitutively, causing fluorescence when the RNA aptamer is in the presence of DFHBI or DFHO.

Constitutive Squash
