C120-trucCYC1p+hbax+ADH1terminator
This Part functions as a blue light-sensitive kill switch when transformed into yeast with BBa_K4238003.
The C120-trucCYC1promoter causes dimeric EL222 to bind to the C120 sequence under blue light, allowing transcription of the downstream sequence. Therefore, hBax is expressed only when the yeast gets exposed to blue light, leading to yeast death. The apoptosis-inducible human Bax gene (BBa_K2365048) was used as the kill switch. hBax has been shown to be able to induce apoptosis in yeast. This kill switch is also expected to be utilized in the field of biosecurity, as apoptosis induces the thin cleavage of genome sequences. Sequence and Features
