MN4_trigger_2_w/oPol

It is a single-stranded DNA sequence having 35 nucleotides. The 5’ and 3’ ends of the sequence have a few bases that are complementary to part Aptamer_MN4 (BBa_K4438800). The middle region has ssDNA sense T7 promoter sequence. Figure 1A) shows the secondary structure and minimum free energy.

Usage and Biology

This part can bind to Aptamer_MN4 (BBa_K4438800) due to complementarity and disrupt its three-way junction. Figure 1D) Shows the secondary structure of both parts hybridised at 37° Celsius. Quinine binds with cocaine-binding aptamer MN4 [1] and displaces the MN4_trigger_2_w/oPol(BBa_K4438803). This part has complete complementarity with MN4_Target_2(BBa_K4438804). T7 polymerase binds and in-vitro transcription of the duplex forms multiple broccoli light-up aptamers. All three parts can be used to detect different concentrations of quinine.
