UM, Monomeric blue chromoprotein
Ultramarine is a blue, monomeric chromoprotein derived from the multimeric Rtms5 chromoprotein (1). Its structure is highly similar to GFP and other chromoproteins, depending on a beta-barrel structure for its function. Its absorbance spectrum shows maximum absorbance at 586nm. The major advantage to using Ultramarine comes from the fact it is monomeric. Other chromoproteins are frequently tetrameric. By using a chromoprotein which does not need to form a tetramer secretion of functional chromoproteins becomes easier for example. A C-terminal 6xHis tag is included to allow for protein purification for colour assays.
We introduced silent mutation at position 423 (C to G) and 519 (C to T) to remove EcorI site and make it Biobrick compatible.
