CrtW: β−carotene ketolase

This part is an Escherichia coli codon optimized version of the ß-carotene-ketolase (CrtW148) from Nostoc punctiforme PCC 73102 (UniProt B2IZ40).

Usage and Biology

The ß-carotene-ketolase (EC 1.14.99.63) catalyses the conversion of ß-carotene into echinenone and of echinenone into canthaxanthin (Figure 1).

The Nostoc punctiforme CrtW [1] is a 30.743 kDa protein of 262 amino acids.

When placed under the control of the hybrid T5 promoter regulated by LacI (BBa_K4432000) or the classical pLac promoter (BBa_R0010), this part leads to the synthesis of canthaxanthin in an E. coli producing ß-carotene.
