Description: This is the sequence for the gene that encodes the GFP binding designed ankyrin repeat protein (DARPin). The DARPin can bind reversibly yet with a very high affinity to the GFP molecule. The affinity of the binding is around 10-30 pM. The GFP DARPin sequence was taken from the NCBI database (NCBI), and can be found with the 5MA6 protein ID. We used the GFP DARPin for test rounds of ribosome display, as well as for collaboration purposes. The GFP protein used as the target was not the exact 5MA6_A protein that was described in the paper (Hansen et al., 2017), however, we confirmed that the small sequence differences between our GFP protein and the paper’s GFP did not matter in this case, by using BLAST.

GFP bindning DARPin

DARPin (designed ankyrin repeat protein) that binds specifically to GFP protein. pdb: molecule 5MA6, Chain B, 3G124nc. Consists of 5 alpha helixes, that bind to the GFP protein.

Characterization: This GFP-binding protein has been expressed as a part of a composite part. Please check out the BBa_K415900 for the details of how we used this basic part in our ribosome display experiments. And BBa_K4159012 for how we used this part for our GFP-DARPin expression cassette.
