The transformation, miniprep and the gel (undigested and digested with the XbaI and pstI) worked. Further tests were confirmed by PCR analysis and subsequent sequencing. The construct can further be characterised with the combination of BBa_I13601 which contians a LacI operator with a CFP reporter. The combination of BBa_K182001 and BBa_I13601 should repress the CFP expression. This inhibition can be removed by the addition of IPTG which will restore the CFP expression in the cell.

Aberdeen_Scotland 2009

The transformation, miniprep and the gel (undigested and digested with the XbaI and pstI) worked. Further tests were confirmed by PCR analysis and subsequent sequencing. The construct can further be characterised with the combination of BBa_I13601 which contians a LacI operator with a CFP reporter. The combination of BBa_K182001 and BBa_I13601 should repress the CFP expression. This inhibition can be removed by the addition of IPTG which will restore the CFP expression in the cell.
