Assembly of Pre B1 (pSB1C3)
The gene fragment Pre B1 is composed of Pre A1 and the TetR gene BBa_C0040. We digested the vector fragment Pre A1 with SpeI and PstI; the insert fragment BBa_C0040 with XbaI and PstI. We ligated these fragments together to form Pre B1.
