For the display of a protein of interest (POI) on the bacterial surface, the sequence encoding for the POI must be inserted between the signal sequence and the coding sequence of fimH. The signal sequence serve as site that is recognised by the secretion machinery and will be cleaved during the secretion process. This results in insertion of the POI in the type 1 fimbriae <sup>1</sup>.

fimH

FimH is the distal protein found at the tip of the type 1 fimbriae.1 The type 1 fimbriae plays a role in the adherence of Salmonella Typhimurium to the host cells, whereby mannose-containing glycoproteins can be bound by FimH.
As a part of our project "Target Taxi", we aim to fuse rBC2LCN (BBa_K3861001), a lectin able to bind specifically to several cancer cell types, to FimH for surface display.
