CBD-SA is the linker module which connects cellulose and aptamer. It is a fusion protein composed of two cellulose-binding-domain and one Streptavidin domain that is produced by engineered Bacillus subtilis. The actual structure of this linker module refers to CBD-SA-CBD. We wanted to use system with B. subtilis to optimize our production of CBD-SA. But, the plasmids of CBD-SA weren’t inoculated into cells of B. subtilis.

In those discussions, however, our team have realized some of the errors that we made, namely, the wrong shaking frequency of the shaker that we set for 200rpm instead of 90 rpm, resulting in the death of the competent cells. Although we have formed plans of solving this problem, our B.subtilis transformation experiment cannot be continue due to the limited time and the used out competent cells.
