Lambert_GA iGEM 2021 developed a cell-free nitrate biosensor, using the pNAR promoter to quantify the amount of nitrate present in a solution. Lambert_GA’s part BBa_K3725220 is a composite part with sfGFP driven by this pNar promoter. The team’s biosensor utilized three plasmids, NarL, NarX and a sfGFP part with the pNAR promoter. These plasmids were designed by Dr. Adam Silverman at the Jewett Lab at Northwestern University and shipped to our lab for testing.

Phosphorylated NarL-Specific Promoter Sequence (pNar)

pNar is a regulatory promoter responsive to phosphorylated NarL. This gene is modeled after the E.coli natural nitrate sensor, the Nar Operon, which includes the membrane-bound sensor protein NarX and its corresponding DNA-binding response regulator NarL.
